AAV
vector production (serotypes 1-6) AAV MSDS
AAV vectors are generated using a triple transfection protocol
and purified using a sequential process of nuclei isolation,
density gradient centrifugation and heparin sulfate affinity
column chromatography.
Vectors are titered by dot-blot and guaranteed to have at least 10E11 total particles. Approximate turnaround time for preps: 3-4 weeks. If you are sending plasmids, please send at least 200 ug.

Adenovirus vector production Adenovirus MSDS
Recombinant Ad vectors are amplified in 293 cells and purified
with 2 consecutive CsCl gradients. Vectors are titered
by infecting target cells and detecting infected cells with
antibody staining. Preps have a guaranteed titer of 10E12 total particles, and approximate turnaround time is 3-4 weeks.

Generation of adenovirus recombinants Adenovirus MSDS
The AdEasy system as described by Vogelstein's laboratory
is used to incorporate new genes into Ad vectors. Cell
lysates are then provided to the investigator for testing
prior to amplification of the vector. Approximate turnaround time: 6 weeks.

Lentivirus vectors Lentivirus MSDS
Recombinant lentiviral vecors are produced in 293T cells by
transfection and are purified/concentrated using sucrose gradients.
Titers of physical vector particles are determined by p24
gag. ELISA. Safety assays for replication competent
retroviruses (RCRs) include marker rescue assay and gag and
tat transfer assays.
The Vector Core offers concentrated preps (100 µl, 10E9 iu/ml) and non-concentrated preps (100ml, 10E6 iu/ml). Approximate turnaround time for concentrated preps: 6-7 weeks; non-concentrated preps: 3-4 weeks.
If you are sending plasmids, please send at least 750 ug concentrated at 1mg/ml. Cesium plasmid preps are recommended, and can be prepared by the Vector Core for an additional $250.
