How to order:

For your convenience, the UNC Vector Core Order Form is available online. 
Please email completed form to Lori Nisi.

March 2013 - We are implementing a new billing system, and the following changes apply:

  • We must receive a PO# or indication of credit card payment on your order form before your order can be entered in our system.
  • For credit card orders, you will now receive an emailed secure link to enter the credit card payment after your order ships (instead of providing the information over the phone).
  • Within the next month, we will transition to an online ordering system.  This will replace the Vector Core order form submission process.  An email will be sent out before the transition.

  • PO payment - Please write PO# on order form.  An invoice will be sent to the accounting contact after order is shipped.

  • Visa/MC - Please indicate you would like to pay with a card on the order form and provide a contact for the card.  You will receive an email with a secure link to enter your credit card information.  We do not charge until your order has been shipped.

  • Shipping Fee:  Vectors are shipped via Fedex.  Shipping charges are $40 per domestic order, $150 per international.

AAV Vector Production (serotypes 1-6, 2.5, 8, 9)   AAV MSDS

Our new AAV vector production method utilizes a triple transfection protocol in suspension HEK293 cells and is purified by column chromotography. This system is serum free, antibiotic free, generates less empty particles, has higher purity and is scalable to Wave bioreactors.  We offer custom vector production and aliquots of in-stock vectors.

Custom AAV Vector Production

For custom vector preps, please send at least 300ug of plasmid.  Approximate turnaround time is 2-3 weeks.  We guarantee a titer of 10E11, although average yield is usually ~10E12.  Yield is dependent on customer construct.

Adenovirus Amplification Prep     Adenovirus MSDS

Recombinant Ad vectors are amplified in 293 cells and purified with 2 consecutive CsCl gradients.  Vectors are titered by infecting target cells and detecting infected cells with antibody staining.  Preps have a guaranteed titer of 10E12 total particles and approximate turnaround time is 3-4 weeks.

Generation of Adenovirus Recombinants      Adenovirus MSDS

The AdEasy system as described by Vogelstein's laboratory is used to incorporate new genes into Ad vectors.  Cell lysates are then provided to the investigator for testing prior to amplification of the vector.  Approximate turnaround time is 6 weeks.

Lentivirus Vectors     Lentivirus MSDS

Recombinant lentiviral vectors are produced in 293T cells by transfection and can be purified/concentrated using sucrose gradients or by size exclusion.  Titers of physical vector particles are determined by p24 gag ELISA.  We translate 1 ng of p24 to 10E4 IU (for most vectors this is a significant underestimation of the actual vector titer).  Safety assays for replication competant retroviruses (RCR) include marker rescue assay and gag and tat transfer assays.  For information on lentivirus production, please contact Dr. Tal Kafri at 919-843-7365.

High Titer Concentrated/Purified Lentivirus Preps (100ul, >10E9 iu/ml)

For in vivo gene delivery studies, the UNC Vector Core recommends using high titer concentrated/purified vectors.  Approximate turnaround time is 6-7 weeks (this includes mandatory safety assays to rule out the emergence of replication competent retrovirus).  Please send at least 350ug of plasmid concentrated at 0.5 - 1.5mg/ml extracted by either Midi/Maxi prep columns or by cesium gradient. Alternatively, plasmid DNA can be prepared by the Vector Core for $250.

High Titer Non-Purified Lentivirus Custom Prep (100ul, 10E8-5x10E9 IU/ml)

Vectors in this category are suitable for in vitro studies which require gene delivery to relatively non-permissive target cells.  Typical vector titers range between 10E9 - 5x10E9 IU/ml (determined by the nature of the vector cassette).  The vectors are concentrated by size exclusion (RCR assays are not mandatory).  A total prep of 100ul can be provided in aliquots of either 10, 20 or 100ul.  Approximate turnaround time is 2-3 weeks.
